چکیده

The distribution of alkylated deoxynucleosides and bases has been determined in the DNA of a sensitive and a resistant human gliomaderived cell line exposed to therapeutic levels of |'"l1|.Y-(2-chloroethyl)/V-nitrosourea in vitro. The resistant cell line is 5-fold less sensitive to the cytotoxic effects of yV-(2-chloroethyl)-Ar-nitrosourea and 8-fold less sensitive to sister chromatid exchange than the sensitive cell line. In comparison with the sensitive cells, DNA from the resistant cells contains much less of the cross-link, l-(3-deoxycytidyl),2-(l-deoxyguanosinyl)ethane. DNA from the resistant cells also contains significantly fewer minor base modifications. The decrease in l-(3-deoxycytidyl),2-(l-deoxyguanosinyl)ethane cross-link formation is probably explained by the higher level of O'-alkyltransferase in the resistant cell line. The lower levels of other DNA modifications could be explained by the presence of higher levels of other DNA repair activities.

Reduced level of DNA cross-links and sister chromatid exchanges in 1,3-bis(2-chloroethyl)-1-nitrosourea-resistant rat brain tumor cells.

We found that 9L-2 cells, a cell line derived from the in vivo 9L rat brain tumor model, are approximately 8-fold more resistant to the cytotoxic effect of 1,3-bis(2-chloroethyl)-1-nitrosourea (BCNU) than are sensitive 9L cells. Treatment with BCNU induces sister chromatid exchanges in both lines, but to produce similar levels of exchanges, 9L-2 cells must be treated with a 14-fold higher conce...

مقایسه روش پرتو درمانی هدفمند با به کارگیری دی‌اکسی یوریدین نشان دار شده با ید ـ 125 با روش پرتو درمانی خارجی، در درمان گلیوما در شرایط

Gliomas comprise about 50% of all primary central nervous system tumors that have defied treatment. Despite of improvement in treatment with surgery, radiotherapy and chemotherapy, the prognosis for these patients remains poor. Efforts to improve the treatment of malignant glioma have included Targeted Radiotherapy with [125I]-Iododeoxyuridine. 125IUdR, a thymidine analogue, is preferen...

DNA interstrand cross-link and free radical formation in a human multidrug-resistant cell line from mitomycin C and its analogues.

A subline of the human breast tumor cell line (MCF-7), selected for resistance to Adriamycin and having the multidrug resistance phenotype, also developed significant cross-resistance to mitomycin C and its two analogues, BMY 25282 and BMY 25067. Because mitomycin C and the analogues contain both quinone and aziridine moieties, the mechanism of tumor cell kill is thought to involve alkylation a...

The Escherichia coli AlkB protein protects human cells against alkylation-induced toxicity.

Escherichia coli can ameliorate the toxic effects of alkylating agents either by preventing DNA alkylation or by repairing DNA alkylation damage. The alkylation-sensitive phenotype of E. coli alkB mutants marks the alkB pathway as an extremely effective defense mechanism against the cytotoxic effects of the SN2, but not the SN1, alkylating agents. Although it is clear that AlkB helps cells to b...
